Protonation et déprotonation dans l'état fondamental et le premier état singulet excité de l'hypéricine en milieu micellaire non ionique

Résumé

L'hypéricine, un pigment naturel, présente une activité antivirale (HIV) induite par la lumière et il a été suggéré que cette activité serait liée à sa capacité à libérer des protons sous l'effet d'une excitation optique. Les valeurs de pKa des équilibres de déprotonation des états singulets fondamental et excité de l'hypéricine en dispersion micellaire (Brij 35) ont été déterminées par titrations spectrophotométriques et spectrofluorimétriques et en utilisant la méthode de calcul de Förster : pKa₁ = 7 ; pKa₂ = 11 ; pKa₁.* = 7 ; pKa₂* = 8,3. Les spectres d'absorption et de fluorescence d'une forme anionique ont été obtenus et le rendement quantique de fluorescence a été mesuré : Φ_F = 0,023. De la même manière les valeurs de pKa des équilibres de protonation ont été aussi déterminés : pKa = 1 ; pKa* = 0,3. Les spectres d'absorption et de fluorescence d'une forme monoprotonée ont été obtenus et le rendement quantique de fluorescence a été mesuré : Φ_F = 0,06.

La protonation et la déprotonation provoquent respectivement un déplacement bathochrome et hypsochrome des spectres. Les deux sites de déprotonation ont été localisés respectivement sur un groupement hydroxyle en position 7 ou 7' (pKa₁) et sur un groupement hydroxyle en position 4,5,4' ou 5' adjacent à un groupement carbonyle (pKa₂). Les propriétés observées ont été interprétées en faisant intervenir un transfert intramoléculaire d'électrons au cours de la première transition Si ← S₀. En conclusion l'hypéricine en dispersion micellaire non ionique n'est pas déprotonée dans le premier état singulet excité.

Abstract

Hypericin, a natural pigment, possesses light induced antiviral activity against the human immunodeficiency virus and it was suggested that this virucidal activity may be related to its ability to acidify its environment upon optical excitation. Ground and excited singlet states déprotonation pKa values of hypericin in water micellar dispersion (Brij 35) were determined by means of spectrophotometric and spectrofluorimetric titrations and Förster cycle calculation : pKa₁ = 7 ; pKa₂ =11; pKa₁* = 7 ; pKa₂* = 8.3. Absorption and fluorescence spectra of an anionic form were recorded and the quantum yield of the fluorescence was measured : Φ_F = 0.023. In the same way ground and excited singlet states protonation pKa values were also determined : pKa = 1 ; pKa* = 0.3. Absorption and fluorescence spectra of the monoprotonated form were recorded and the quantum yield of the fluorescence was measured : Φ_F = 0.06

Protonation and déprotonation induced respectively a bathochromic and a hypsochromic shift of the spectra. The two sites of déprotonation were deduced by comparison between spectra of the anionic form and spectra of hypericin-met al complexes, to be respectively the hydroxyl group in position 7 or 7' (pKa₁) and the hydroxyl group in position 4,5,4' or 5' with adjacent carbonyl group (pKa₂). Electron donor groups such as -OH or -O^- having lone electron pairs which are partially conjugated with the aromatic ring system, thus the first transition Si ← S_o is also of the intramolecular CT type, with the functional groups being more extensively conjugated with the aromatic ring and therefore more charge deficient in the lowest excited singlet state than in the ground state It was concluded that hypericin in non ionic micellar dispersion is not deprotonated in the first excited singlet state.
